Son abréviation officielle d'auteur botanique est Orb. Il était le frère du naturaliste Alcide Dessalines d'Orbigny, fit ses études à La Rochelle et étudia la médecine à Paris. En 1834, il succède à Louis Cordier (1777-1861) comme assistant et préparateur de géologie au Muséum national d'histoire naturelle de Paris. Lors de la création de la Société Cuvierienne en 1838, il était l'un des 140 membres fondateurs de la société. D'Orbigny est rédacteur en chef de l'encyclopédie d'histoir.
